Well, to gain access to the said chest you have to have completed Chapter 2 of the Seasonal Journey. Many have overlooked this part of the quest requirements, so once you have completed Chapter 2 of the Seasonal Journey, go back to the chest and you will be able to open it.

How To Complete Chapter 2 in Diablo 4 Season of Blood
To do so, first, you must have completed/skipped the campaign, and then you must complete these tasks:
- Forsaken Offerings: Obtain 2 Caches from the Tree of Whispers by completing Whispers.
- Vault Delver: Complete 8 Cellars.
- Ruins Reclaimed: Conquer a Stronghold.
- Finer Clarity: Upgrade your Potion to Minor at the Alchemist.
- Path to Strength: Unlock 8 Aspects in the Codex of Power
- Baptized in Blood: Reach Level 25.
- Deliver Us From Evil: Rescue 5 Villagers within the Blood Harvest
- Vampire No More: Defeat a Blood Seeker
- Tides of Blood: Activate a Vampiric Power that requires at least 3 Pacts
- Contract Reforged: Add or remove Pacts from your armor
- Bejeweled: Craft a Gem at the Jeweler.
You must complete at least nine of these 11 tasks for the Chapter to be considered complete.
When you have finished this, simply return to the Fetid Mausoleum dungeon with Hunter Erys, open the chest, retrieve the Profane Vampire Skull, and continue on with the questline.
