I think 360 would have the edge. Many more gamers that play online use headsets with the 360 and communicate more to win matches.
Also the Dualshock2 lags way behind the 360 controller when it comes to long gaming sessions - it's designed for lobsters not humans.
Pick up a 360 controller and your fingers automatically sit on the triggers, your thumbs naturally find their way to the analogue sticks. You don't feel this with the Dualshock.

Maybe this isn't the best example of it in action - games where lots of individual levels load when you jump from one level to another it will help more. Doesn't GTA basically do one big data load then streams it afterwards?
That said, I'm not actually bothered about load times, the noise aspect is why I will install games to the HD!
Not sure co-op really lends itself to the Mass Effect series - aside from combat, are the other 4 players just going to sit there and listen to the conversations chosen by only one player? Talking is a pretty big part of the game.

I'm a die-hard Xbox fan but I'll be buying a PS3 soon. Main reason? Not games, but to use it as a media centre (& BluRay) now that I've bought a HD TV. The xbox is simply too loud to watch movies on especially late at night.
I'm not saying that it makes you miraculously better at gaming, but the peripheral you're using definitely can make give you the edge in online matches. Why else do companies sell optical mice with higher dpi, programmable buttons etc specifically for gaming? It doesn't make them a better player, but it might make a split second difference which will give them the edge.
