Not only will GTA V run on a single DVD, Xbox 360 owners will also receive access to DLC first. Look for an announcement this December at the Spike VGA's.
Let's not forget, GTA 4 ran better on the 360 (PS3 was riddled with framerate drops as many reviews pointed out) and also gave 360 owners the Lost and Damned and the Ballad of Gay Tony over a YEAR before PS3 owners received it.
