In fairness, we shouldn't we joke about rape, it contributes to an overall cultural trivialization of the subject. As if people, in their fear of it as a reality to be dealt with, decide instead to joke about it- to "pad" it with humour. Humour as a distancing tool.
I know this is just one instance, but #1. He's an authoritative voice in the videogames industry and #2. People make these kinds of tasteless jokes all the time; it's hard to attack everyone all at once about it. To be honest I think admonishing a "famous" person about it is more effective than admonishing some random joe public. It makes waves like this is sort of doing.
The thing is though, he didn't even mean it in the sense that Jeff Gertsman was going to rape him. He meant that he looked tired, something that roofies do to you and because it is used as "the date rape drug" people assumed he was making light of rape. I bet rape didn't even cross his mind when he tweeted that.
You think it's OK to tarnish the reputation of a famous person just to bring a subject to light? What does a person's social standing have anything to do with misrepresenting a person's character? You think it's OK to make an example of someone famous just to push an agenda? How would you feel if you were in that situation, famous or not? Ken Levine, while popular, is still a human being, and as such he deserves to behave and be treated as one just like anyone else.
I'm sorry, but in the instance of rape, I think the mainstream media and most people I know pretty much think it's reprehensible. I don't know why you have to drag someone's name through the mud just to make a point that really doesn't need to be made.
I say many many inappropriate things in the company of friends, and they say them right back. We understand it's a joke, and believe me if anyone of us were faced with a terrible thing such as rape, sexism, racism, assault, or whatever we would handle that situation with all seriousness and not make jokes about it.
We're adults, and as such we can separate reality from just having a good laugh.

This whole thing was taken out of context without even knowing the man. For all we know being roofied is just a colloquialism for "someone took me out and got me piss ass drunk the night before". I say this, because I've heard it used this way on more than one occasion. From his original comment, the only rape implied was by those accusing him of inappropriateness.
Re: everyone thinks rape is reprehensible... Yeah, I dunno. People say they're against rape, until it happens. Then it's the victim's fault for being in a situation where they could be raped.
Raped in India? Well, why were you there, you know it's dangerous, you shouldn't have been outdoors at night. Raped by football players? Well, why'd you get so drunk and ruin their lives (seriously, CNN kind of said this)? Raped by your husband? Well, if you didn't want it, why did you marry him, or stay with him? Raped while being a man? You must have been in prison and deserved it, or weren't a real man in the first place. Raped while young? You probably wanted it, and you're only talking about it now because someone found out. And on and on... it's really sad.
On the actual topic, though, I don't see this tweet as such a big deal. Probably only became one because he's got a lot of followers, and statistically some of them have been roofied and raped, so it's hard for them to hear roofies spoken of casually. It's why people who are high profile should generally just be careful about what they say.
Rape has happened in my family, really close to it and I didn't even think of it until people pointed it out. Unless you have even dealt with this issue first hand, whats give you the right to say anything on the matter?

A roofie is a pill that basically knocks a person out; some shady men use it to knock out a women at a bar by spiking her drink with it and rape them.
