The Red9 Handgun cannot be obtained until Chapter 4 of Resident Evil 4 Remake, when you can access the boat and explore the lake. Once you’re on the boat, head towards to center of the lake. You’ll know when you’re at the center because you’ll see a shipwreck.

Is the Red9 Missable in RE4?
