Runes in the Lost Temple is triggered by finding any rune in the Lost Temple of Dirthamen. You'll need one member of all three classes to get all the runes on one pass.
When you first enter the area, go forward (north) while keeping your eyes right. You'll see a Veilfire Brazier near the corner of the stone at the water's edge. The brazier will be marked on your map. Light a Veilfire Torch with it, then face north. You'll see a stone shaped similarly to a headstone. Examine the rune on it while holding the Veilfire Torch. This triggers the quest.
