ND said that Joel's and Ellie's story is done, so I think the DLC will be related to either Tess or maybe the Fireflies. It would be cool to see how Joel became a smuggler and maybe his first struggles
I remember Neil Druckman said that he meant Joel and Ellies story is done in terms of them trying to get a cure. He didnt mean for them to go away I think we are stil going to get these characters in a sequel I just dont see the games without them.
He said that he was misquoted and their story isn't finished. In a Q&A he had with fans, he mentioned that Ellie and Joel could return for a sequel. I would much prefer they returned to Ellie and Joel then to have new characters. They're such great characters to just leave alone, especially Ellie.
Maybe something to do with Ish (could have cutscenes of him writing the letters that you find when you play as Joel), or maybe Sam and Benjamin. I personally would like to see DLC as a Younger Joel with his brother not long after the outbreak.
I hope it is Ish, but it could be anything. Might play as Bill :D
This is one of the big aspects that makes the game so great, the characters. If they took the route of following a character, there are plenty to choose from. For me I would love to see a David story or more Bill (Can't have enough Bill).
Can't believe i forgot about Bill. Damn, i hope they do more than just one piece of singleplayer DLC. There are too many great opportunities to only do one.

I'd love to see Sam and Henry's story just prior to them Meeting Joel and Ellie. Or as above, Ish's story.

I always wanted to see what happened during those 20 years that were skipped. How does Joel meet Tess? Why is Tess important? How does the outbreak spread so rapidly? etc.... They could answer some questions and let us see what happened without ruining the ending. In fact, they may strengthen it.
I would really like to see Marlene in the DLC. I think her journey would be awesome and possibly with the help of an AI dog as a companion. Either that or new character.
